We investigated whether concurrent ingestion of dengue 2 virus and microfilariae of Brugia malayi would increase viral infection and dissemination rates in Aedes aegypti. Infection rates were similar in mosquitoes that ingested virus along or both virus and microfilariae concurrently. However, viral dissemination rates, as determined by recovery of dengue virus from both legs and bodies separately, were significantly greater in mosquitoes that ingested both agents concurrently than in those...

A microhematocrit tube technique for diagnosis of human filariasis has been previously described. A system incorporating heparin, EDTA (ethylenediaminotetraacetate) and acridine orange into a microhematocrit tube (Quantitative Blood Count, QBC) has been commercially developed for the quantitation of blood counts and has been used for the diagnosis of malaria. We evaluated this test for its usefulness in the diagnosis of filariasis. Upon centrifugation, the parasites were concentrated in the...

A study has been made of the distribution of nematode plant parasites and of the damage they cause in all branches of plant breeding. During the past few years, Soviet prognostic centers have begun to reckon with nematode diseases. Since 1963 periodic symposia have been published to serve as brief summations of research conducted on parasitic nematode distribution and damage in the USSR.

A survey of macroinvertebrates and their habitat was conducted in Luxapalila Creek soon after completion of flood control measures in the lower reach near Columbus, MS, 1998-2000. The results of this study were compared with a similar preconstruction survey conducted in 1987-1989. There was no evidence of adverse effects of construction on the benthic macroinvertebrate community. Recently channelized, unchannelized, and historically channelized portions of the stream differed somewhat in...

The pathogenicity of the entomogenous nematodes, Neoaplectana carpocapsae (DD-136 strain) and Neoaoplectana glaseri, was tested for the German cockroach, Blattella germanica, under laboratory conditions. All developmental stages, including oothecae, were tested for susceptibility. Infection dishes consisted of standard size petri dishes lined with filter paper, to which were added 3 ml of a nematode suspension containing infective juveniles at a concentration of 1000, 5000, 10,000 and 20,000...

A gall is formed not as a result of the influence of the basic growth hormones which originate in the plant itself, but either under the influence of the toxins excreted by the nematode or, more preferably, of the products of the interaction of the nematode with the plant. These toxins also change the normal growth processes. It is known (8) that several amino acids stimulate the pathological growth of meristem. It is possible also that the liberation of amino acids by the nematodes is a reason...

With the goal of developing a biological assay method for staphylococcal enterotoxin, the effect on the development of nematodes of the order 'Strongyloides', Rhabditis oxycerca de Man and Panagrellus redivivus Goody was examined. After preparation by fasting and treatment with 0.02 - 0.2M Na2HPO4.12 H2O, 750 - 2,250 units per ml of Hylase, or 1 - 5 mg per ml bacterial protease, or with untreated mixed cultures (adults and larvae), significant resistance was demonstrated in the case of both...

Apoptosis is a major determinant in the ability of cells to become cancerous. The aim of this grant was to identify conserved regulators of apoptosis using the nematode C. elegans as a model system. Specifically, we searched for proteins that interact with CED-9, the nematode homolog of the oncoprotein Bcl-2. We found that CED-9 interacts with CED-4, the worm homolog of the apoptotic regulator Apaf- 1, and characterised the regulation of CED-9 by EGL-1, a BH3 domain protein that also directly...

The islands of Batan and Rapu Rapu in southeastern Luzon, Philippines, were surveyed for filariasis from November 1984 to May 1985. Microfilariae of Bancroftian filariasis were detected in 10.5% of the people over one year of age. Microfilarial rates and intensity of infection were the same for males and females. The highest rates for both sexes were in the 56- to 60-year-old age group. Aedes poicilius, which breeds in abaca and banana plants, was found to be very anthropophilic, comprising...

The main goal of our grant application is to identify novel genes that function in apoptosis (programmed cell death), using the C. elegans cell death suppressor CED-9 as our starting point for an extensive yeast two-hybrid screen. In the first year of funding, we have concentrated our attention on our discovery of a specific interaction between CED-9 and CED-4, one of the proapoptotic proteins in C. elegans. We showed that CED-4 is likely to be a major target for CED-9 action. We have also...

Approximately one thosand vials of preserved nematodes, collected from fishes, amphibians, reptiles, birds and mammals were studied by conventional parasitological techniques. Species new to science were described and named. Poorly known species were redescribed, often with revision of the higher categories of classification and with identification keys to the genera and species. New host and distribution records were recorded, and those parasites with obvious potential for infecting man were...

The lab has grown significantly in the past year. Two postdoctoral fellows mentioned in the previous Progress Report are now in house and fully up to speed. One is working on the physiological aspects of the project (Specific Aim 2); the other is working on the modeling (Specific Aim 3). I am also pleased to report that I now have a graduate student. He is working exclusively on the behavioral analysis of nematode chemotaxis. I now have, therefore, implemented my original plan for the lab: to...

The influence of diesel fuel on benthic food webs in coastal salt marshes were examined in a series of microcosm experiments. Diesel contamination resulted in high mortality to all copepods except Cletocamptus deitersi, while nematode abundance increased. Blooms of benthic microalgae that developed in contaminated sediment were primarily a response to reduced copepod grazing, and secondarily a consequence of enhanced NH4+ flux from sediments. Microalgal grazing by nematodes and copepods was...

When virus and microfilariae are ingested concurrently by a mosquito, microfilariae (mf) may penetrate the mosquito midgut and introduce virus directly into the mosquito hemocoel, allowing mosquitoes to become infectious much sooner than normal and enhancing transmission of viruses by mosquitoes. Mansonella ozzardi (Manson) is a benign filarial nematode parasite of humans in Latin America and is transmitted by black flies (Diptera: Simuliidae) and biting midges (Diptera: Ceratopogonidae)....

Genetic experiments in C. elegans have shown that a triad of genes consisting of CED3, CED4, and CED9 is necessary for all 131 apoptotic events occurring during the development of the nematode. Homologues of these genes have been identified in mammals as the caspase Family, Apaf 1, and the Bc1 2 family. Targeted gene studies have shown that most caspases involved in apoptosis have a non redundant function. Apaf4 also seems to be involved in most apoptotic event& On the other hand, the mild...

The extent to which plant diseases and parasites occur varies with the habitats, and for this reason the yield of certain crops grown on these different habitats is endangered by these damage-causing agents to a greater or lesser extent. In the case of one-crop agriculture these dangers will appear sooner or later. This observation very early determined the course of action adopted by the farmer and gave rise not only to the form of agriculture in which different parcels of land are assigned to...

The pathogenicity of two different entomogenous nematodes (Neoaplectana carpocapsae and N. glaseri) for the German cockroach (Blattella germanica) was investigated. Infective juveniles of each test species were placed in an infection chamber consisting of a standard petri dish with filter paper. At sufficient concentrations, the invasive of N. carpocapsae (DD-136) reduced roach populations significantly. However, the invasive of N. glaseri proved considerably less virulent.

We have examined and documented several changes in the benthic community structure of Lake Pontchartrain. The change from dominance by large Rangia cuneata to dominance by very small hydrobiid gastropods, for instance, has occurred since the last large-scale study was done 25 years ago. We have also discussed changes in benthic community function in Lake Pontchartrain. We have characterized the lake as a functioning as a one-resource system, with the filter-feeding zooplankton, the...
