Structural designers today face both opportunities and challenges in a vast array of available analysis and optimization programs. Some programs such as NASTRAN, are very general, permitting the designer to model any structure, to any degree of accuracy, but often at a higher computational cost. Additionally, such general procedures often do not allow easy implementation of all constraints of interest to the designer. Other programs, based on algebraic expressions used by designers one...

In October 1997, the Princess Royal opened the Wellcome Trust's Human Genome Project Research Centre at Hinxton near Cambridge. The campus is also the seat of the European Bioinformatics Institute, the Sanger Centre of the Wellcome Trust, and the MRC's Human Genome Project Mapping Resource Centre. The aim is to decode more than one-sixth of the human genes by the year 2002. This short film explains some of the work that goes on at Hinxton. 1 segment.

In comparison with that known from later geologic time, the Archean fossil record is miniscule: although literally hundreds of Proterozoic formations, containing more that 2800 occurrences of bona fide microfossils are now known, fewer than 30 units containing some 43 categories of putative microfossils (the vast majority of which are of questionable authenticity) have been reported from the Archean. Among the oldest known fossils are Early Archean filaments reported from cherts of the Towers...

The bones of the feet are likely to lose mineral during spaceflight because they are not functioning to support body weight in space. Although loss of bone from the heel bone (calcaneus) during spaceflight is well documented, loss of mineral from other bones in the feet of astronauts has not been quantified. A possibly vulnerable candidate is the metatarsal, which, as a weight-bearing bone in runners, is vu I nerable to stress fractures. To explore the potential for bones of the feet to lose...

Electro-Optic Polymers hold great promise in increased electro-optic coefficients as compared to their inorganic corollaries. Many researchers have focused on quantum chemistry to describe how the dipoles respond to temperature and electric fields. Much work has also been done for single layer films to confirm these results. For optical applications, waveguide structures are utilized to guide the optical waves in 3 layer stacks. Electrode poling is the only practical poling method for these...

The principles upon which the process of protein synthesis and the genetic code were established are elucidated. Extensive work on nuclear magnetic resonance studies of both monomermonomer and monoamino acid polynucleotide interactions is included. A new method of general utility for studying any amino acid interacting with any polynucleotide was developed. This system involves the use of methyl esters of amino acids interacting with polynucleotides.

The After the Genome meetings were started in 1995 to help the biological community think about and prepare for the changes in biological research in the face of genomic information. This workshop brings together intellectuals from subject fields far outside of conventional biology with the expectation that this will help focus thinking beyond the immediate future. Hence the subtitle for this year's meeting: "Envisioning Biology in the Year 2010". Accordingly, the organizers brought...

The idea behind genetic algorithms is to extract optimization strategies nature uses successfully - known as Darwinian Evolution - and transform them for application in mathematical optimization theory to find the global optimum in a defined phase space. One could imagine a population of individual 'explorers' sent into the optimization phase-space. Each explorer is defined by its genes, what means, its position inside the phase-space is coded in his genes. Every explorer has the duty to find a...

To support the exploration of space, a reusable space-based rocket engine must be developed. This engine must sustain superior operability and man-rated levels of reliability over several missions with limited maintenance or inspection between flights. To meet these requirements, an expander cycle engine incorporating a highly capable control and health monitoring system is planned. Alternatives for the functional organization and the implementation architecture of the engine's monitoring and...

The available evidence permits one to conclude that streams flowed and lakes developed on Mars sometime in the remote past. The lessons learned from the Earth's earliest fossil record suggest that stromatolites might have formed on Mars, speculating that: (1) biopoesis occurred on Mars during its earliest history; (2) life evolved and diversified; (3) life inhabited aqueous environments; and (4) sunlight was an important environmental resource. The most likely place to find stromatolites and...

The Evolvable Neural Software System (ENSS) is composed of sets of Neural Basis Functions (NBFs), which can be totally autonomously created and removed according to the changing needs and requirements of the software system. The resulting structure is both hierarchical and self-similar in that a given set of NBFs may have a ruler NBF, which in turn communicates with other sets of NBFs. These sets of NBFs may function as nodes to a ruler node, which are also NBF constructs. In this manner, the...

Along present day coast lines, the environmental distributions of prokaryotic and protistan populations are often sharply delimited. Realized habitat ranges are generally narrower than those circumscribed by physiological tolerances, suggesting the importance of organism-organism interactions in the determination of population distributions. Microfossil populations preserved in silicified carbonates, dolomites, and shales of the 700-800 Ma old Akademikerbreen Group, Svalbard, and elsewhere...

Cell-free protein synthesis has emerged as a powerful approach for expanding the range of genetically encoded chemistry into proteins. Unfortunately, efforts to site-specifically incorporate multiple non-canonical amino acids into proteins using crude extract-based cell-free systems have been limited by release factor 1 competition. Here we address this limitation by establishing a bacterial cell-free protein synthesis platform based on genomically recoded Escherichia coli lacking release...

It is generally believed that an RNA World existed at an early stage in the history of life. During this early period, RNA molecules are seen to be potentially involved in both catalysis and the storage of genetic information. It is widely believed that this RNA World was extensive and therefore a sophisticated nucleic acid replication machinery would presumably predate the translation machinery which would not be needed until later stages in the development of life. This view of an extended...

Data bases built from the source literature are plagued by problems of data quality. Unless the data acquisition is done by experts, working slowly, the data base may contain so much garbage that true signals and patterns cannot be detected. On the other hand, high quality data bases develop so slowly that satisfactory statistical analysis may never be possible due to the small sample sizes. Results of a test are presented of the opposite strategy: rapid data acquisition by non-experts with...

The degeneracy of the genetic code allows nucleic acids to encode amino acid identity as well as noncoding information for gene regulation and genome maintenance. The rare arginine codons AGA and AGG (AGR) present a case study in codon choice, with AGRs encoding important transcriptional and translational properties distinct from the other synonymous alternatives (CGN). We created a strain of Escherichia coli with all 123 instances of AGR codons removed from all essential genes. We readily...

In order to understand the structure of real structure spaces, we are studying the 5S rRNA structure space experimentally. A plasmid containing a synthetic 5S rRNA gene, two rRNA promoters, and transcription terminators has been assembled. Assays are conducted to determine if the foreign 5S rRNA is expressed, and to see whether or not it is incorporated into ribosomes. Evolutionary competition is used to determine the relative fitness of strains containing the foreign 5S rRNA and a control 5S...

Much of our recent work has been a study of aminoacyl AMP derivatives. Elucidation of the character of aminoacyl AMP derivatives has made it obvious that AMP has characteristics which should allow it to preferentially catalyze the synthesis of L-amino acid peptides. The essential features which lead to this conclusion are that all l-amino acids (but not all D amino acids) when esterified to 5'-AMP preferentially (65 percent) distribute to the 3' position of the 5'-AMP; that esterification is...

The purpose of this project is to develop a novel cell-based assay that is designed to identify compounds for improving mitochondrial function and metabolic disease. The assay utilizes a dual luciferase reporter system to assess translation regulated by 5-UTR sequences. During the reporting period, the originally designed assay was implemented, validated, and optimized successfully. A small compound library (80 compounds) was tested to demonstrate the utility of the assay for phenotypic...
