The primary objective of this project was to provide the first experimental test of OT's possible causal role in the disinhibition of aggressive behavior in humans. This research was designed to investigate a likely hormonal link between a neurotransmitter implicated in coalition-building and increased willingness to engage in aggressive behavior. Potential applications of this research include increasing our ability to predict and influence aggressiveness by better understanding the...

The purpose of this study is to evaluate the efficacy of intranasal oxytocin on bladder nociception in a cohort of patients with interstitial cystitis (IC). We hypothesize that patients with IC are more likely to experience treatment responses as defined by global response assessment (GRA) scores when they receive intranasal oxytocin as opposed to when they receive intranasal saline. Secondary outcome measures include a verbal pain report (VPR; 0-10 with 0 being no pain and 10 being the worst...

The effects of Coriolis forces on compressible convection are studied using three-dimensional numerical simulations carried out within a local modified f-plane model. The physics is simplified by considering a perfect gas occupying a rectilinear domain placed tangentially to a rotating sphere at various latitudes, through which a destabilizing heat flux is driven. The resulting convection is considered for a range of Rayleigh, Taylor, and Prandtl (and thus Rossby) numbers, evaluating conditions...

The purpose of the grant is to test whether exogenous oxytocin acts as an antianxiety agent and whether social support facilitates its antianxiety effects in a fear-potentiated startle paradigm. Oxytocin given systemically (0.1 micrograms/kg, sc) effectively reduced background anxiety, but not specific cue-potentiated fear. This was found when oxytocin was given either before fear conditioning (acquisition), immediately after fear conditioning (consolidation), or before retrieval/expression of...

The primary objectives of this clinical study are test the hypotheses that (1) cognitive behavioral therapy (CBT) aimed at core social dysfunctions, and (2) oxytocin (OT) administration prior to CBT sessions will each enhance social function in young adults with autism spectrum disorders (ASD), and to examine whether neuroimaging measures of brain function and structure can predict CBT treatment response. To examine these questions, we will recruit and carefully characterize 150 men, ages...

A study of numerical attributes peculiar to an overset grid approach to unsteady aerodynamics prediction is presented. Attention is focused on the effect of spatial error associated with interpolation of intergrid boundary conditions and temporal error associated with explicit update of intergrid boundary points on overall solution accuracy. A set of numerical experiments are used to verify whether or not the use of simple interpolation for intergrid boundary conditions degrades the formal...

Wetlands are important for their role in global climate as a source of methane and other reduced trace gases. As part of an effort to determine whether radar is suitable for wetland vegetation monitoring, we have studied the dependence of microwave backscatter and attenuation on leaf area index (LAI) for flooded rice fields. We find that the radar return from a flooded rice field does show dependence on LAI. In particular, the C-band VV cross section per unit area decreases with increasing LAI....

Organophosphate esters were studied in terms of their ability to affect neuropeptides in the rat central nervous system. The in vivo biosynthesis of enkephalin peptides in the basal ganglia and of vasopressin and oxytocin in the hypothalamus were studied and the effect of subcutaneous administration of diisopropylfluorophosphate (DFP) in the biosynthesis of these neuropeptides was assessed. It was found that DFP inhibited the biosynthesis of vasopressin and oxytocin. These results suggest that...

The object of this study was to determine whether prostaglandin E2, (PGE2) can prevent ovariectomy induced cancellous bone loss. Thirty-five 3-month-old female Sprague-Dawley rats were divided into two groups. The rats in the first group were ovariectomized (OVX) while the others received sham operation (sham-OVX). The OVX group was further divided into three treatment groups. The daily doses for the three groups were 0,1 and 6 mg PGE2/kg for 90 days. Bone histomorphometric analyses were...

The project aimed to identify the neural basis of social behaviors in rhesus macaques, and characterize the role of a set of brain areas in the prefrontal cortex as well as the role of oxytocin (OT) in social behaviors. We have developed an animal model of social interactions and studied the role of specific brain areas in social behavior. We found that the anterior cingulate gyrus (ACCg) is important for signaling reward experience of others and is involved in social learning when monkeys...

Several problems of interest in planetary infrared remote sensing are investigated using a new radiative-conductive model of energy transfer in regoliths: the solid-state greenhouse effect, thermal beaming, and reststrahlen spectra. The results of the analysis are as follows: (1) The solid-state greenhouse effect is self-limiting to a rise of a few tens of degrees in bodies of the outer solar system. (2) Non-Lambertian directional emissivity can account for only about 20% of the observed...

In this issue of Critical Care Medicine, Guarido et al (1) present their fascinating work in a model of endotoxemia in rats. Consistent with prior studies, they found that vasopressin could increase blood pressure in animals refractory to phenylephrine. What is provocative about this work is the presumptive mechanism for these findings. In septic animals, this improvement in blood pressure could not be explained by improvements in cardiac function or vasoconstriction from large vessels. The...

Efforts were directed towards maintenance of actively secreting human pituitary growth hormone cells (somatotrophs) in vitro. The production of human growth hormone (hGH) by this means would be of benefit for the treatment of certain human hypopituitary diseases such as dwarfism. One of the primary approaches was the testing of agents which may logically be expected to increase hGH release. The progress towards this goal is summarized. Results from preliminary experiments dealing with...

Understanding the specific mechanisms involved in the regulation of water balance by the kidney will provide information needed to design preventive measures for dealing with potential adverse conditions that may result in water deprivation and decreased renal responsiveness to vasopressin. The present study is designed to determine the cellular mechanisms involved in reduced fluid reabsorption and membrane reorganization following the removal of the actions of ADH.

The results of this study show that the viability of ova obtained from hormone-treated females is equal to that of ova from spontaneous ovulations in the two strains of mice studied (Swiss-Webster and C57BAL/6Cum). Approximately 90% of all cultured ova developed from the late two-cell or early four-cell stages to blastocysts. The use of exogenous hormones to induce ovulation seems to afford a reliable, efficient, and economic means of obtaining fertilized mammalian ova.

In spite of the fact that a vast majority of the electrophoresis effort (approximately 90%) could not be done on this mission (IML-2) due to failure of FFEU hardware, we find some interesting differences in flight samples obtained from other parts of the experiment. Each part describes the data collected thus far from each of the 4 cell culture kits (CCK) which flew in space. Each CCK was...

Currently, there are no established pharmaceutical strategies that effectively treat core autism spectrum disorder (ASD) symptoms, including pervasive social deficits and repetitive behaviors. The oxytocin pathway has an important role in normal human social behaviors, and oxytocin dysregulation has been implicated in ASD-associated behavioral symptoms. There is now emerging evidence that oxytocin has therapeutic efficacy in ameliorating core ASD symptoms associated with social behavior....

A study of the effects of amino acids on hypothalamo-pituitary capacity to secrete ACTH, and the response of the adrenal gland in terms of corticosterone level in the plasma was discussed in the first part of this report. Second, the report includes a study of the response of protein metabolism in various levels of corticosterone elevation in the plasma. This second part is regarded as having considerable significance for space travel, since this data indicate a threshold level of plasma...

Experiments were carried out to elucidate the role of vasoprressin in the thyrotropin discharge and thyroid secretion, measuring thyroidal 131I uptake (T/S), plasma PB131I level, as well as histologi cal examination of the thyroid gland, in rats. Though a single dose of Pitressin failed to change the T/S in rats kept at 20 degrees centigrade, successive administrations of Pitressin tannate resulted in a suppression of the rise of T/S due to chronic cold exposThese results well coincided with...

The role played by advanced technology is illustrated with respect to the anticipated era of discovery and exploration (in space): how bold new exploration initiatives may or may not be enabled. Enabling technology makes the mission feasible. To be truly enabling, however, the technology must not only render the proposed mission technically feasible, but also make it viable economically; that is, low enough in cost (relative to the economy supporting it) that urgent national need is not...

Pseudotumor cerebri (PTC) is frequently associated with an empty sella turcica. While commonly asymptomatic, it may present with non-specific neurologic symptoms or more rarely with pituitary dysfunction. This case highlights a patient with PTC and secondary empty sella syndrome whose symptoms of hypopituitarism were wrongly attributed to complications in the management of his PTC resulting in delayed diagnosis and management.
